Millbrook was the tobacco estate of John Wayles Eppes in Buckingham County, Virginia. After his wife, Maria Jefferson Eppes (Thomas Jefferson's younger daughter), died, he sold his Eppington plantation in 1810 and moved to Millbrook by 1811. Moving to Buckingham County was politically strategic: Thomas Jefferson wanted his son-in-law to run for Congress to oust a political nemesis, John Randolph. Wayles succeeded in doing this in the 1813 election. It was at Millbrook that Eppes later married his second wife, Martha Burkes Jones.

The estate had a two-story house, which burned in 1866.
